[摘要]目的: 探讨肌钙蛋白在增生性瘢痕形成与发展中所起的作用及意义。方法:收集患者不同时期增生性与非增生性瘢痕作为研究对象,利用肌钙蛋白的基因特异片段,制成寡核苷酸探针与瘢痕组织切片原位杂交,同时,将各标本进行组织块培养,制成成纤维细胞爬片,进行原位杂交,分析其变化规律。结果:肌钙蛋白基因在早期增生性瘢痕中表达均明显强于晚期增生性及非增生性瘢痕。结论:瘢痕增生挛缩与细胞骨架运动的相关基因存在密切关系,而肌钙蛋白起到十分重要的作用。同时,抑制该基因的表达,可能实现减轻瘢痕增生与挛缩。

Effect of gene expression of fibroblast Osteonectin on the hypertrophic and non-hypertrophic scar

[Abstract]Objective:To investigate the effect of gene expression of fibroblast Osteonectin on the formation and contraction of hypertrophic scar.Methods:Osteonectin, one of the most important genes in extracellular matrix, was selected and made into oligonucleotide probe. 24 cases of hypertrophic scar and 24 non-hypertrophic scar and 12 normal skin were used,and these scar were taken on 3, 6, 9, 12 months after burned. Freezing slices and cultured fibroblasts were made to detect the expression of these genes by hybridization in situ.Results:Osteonectin were found the expression in scar tissue, but those in hypertrophic scar were significantly stronger than non-hypertrophic scar.Conclusion:Over-expression of Osteonectin genes act very important functions on the formation of hypertrophic scar.
